An experimental intraradicular biofilm model in the pig for evaluating irrigation techniques

Abstract Background We established an in vivo intraradicular biofilm model of apical periodontitis pigs which we compared the efficacy different irrigant activation techniques for removal. Methods Twenty roots from deciduous mandibular second premolar 5 male were used. After pulpectomy, canals left open 2 weeks and then sealed 4 to enable development intracanal biofilm. The biofilms was evaluated using SEM bacterial 16S rRNA gene-sequencing. To investigate removal, root canal irrigations performed conventional needle, passive ultrasonic, subsonic, or laser-activated irrigation. Real-time PCR conducted quantitate remaining components. Statistical analysis ANOVA followed by a Tukey kramer post-hoc test with ? = 0.05. Results pulp exposure effective inducing revealed multi-layer formation inside canal. sequence identified Firmicutes, Bacteroidetes, Fusobacteria as predominant phyla components, is similar microbiome profile seen humans. None tested irrigation completely eradicated components canal, but subsonic methods produced lowest counts ( p < 0.05). Conclusions An experimental has been successfully pigs.
